Maximilian Kilman's 10th minute goal had the Hammers' season underway perfectly after he got on the end of a Jarrod Bowen corner.
Both teams traded blows throughout the first half, before Burnley managed to have most of the momentum throughout the second period.
However, against the run of play, a clever run by Mahamdou Kante led to him assisting Manor Solomon for a debut goal in the 72nd minute to seemingly set Nuno Espirito Santo's men on their way to victory.
Ollie Scales had other ideas, though, as nine minutes later Zian Flemming scored from the spot after Scarles stupidly brought Marcus Edwards down in the box.
Flemming then completed Burnley's comeback in the second minute of extra time after the Irons failed to clear a corner kick.
Manor Solomon shines on West Ham debut
It wasn't just getting a goal that impressed West Ham fans in Solomon's debut.
Throughout the match, he proved to be a thorn in Burnley's side, offering the Hammers an attacking output when they needed to get up the park.
According to stats by Sofascore, the former Tottenham attacker completed 92 per cent of his passes at Turf Moor.
He also completed 100 per cent of his three dribbles and even chipped in defensively by winning six of his eight ground duels and making one tackle.
Ollie Scarles has shocker in West Ham opening game
At the opposite end of the spectrum to Solomon's performance was that of Scarles.
Although the 20-year-old made one clearance on the line to deny Burnley an equaliser when West Ham were only 1-0 up, the full-back's performance was no more than a great advert for why El Hajdi Malick Diouf needs to stay at the London Stadium.
Scarles conceded a ridiculous penalty in the late stages of the game, lunging in on Edwards and not getting anywhere near the ball.
Call it inexperience if you want, but it was sheer stupidity when the Hammers were two goals to the good and it opened the door for their successful late comeback.
Shortly after that, he saw a West Ham throw-in given to Burnley in the attacking third for taking too long to take it and Nuno promptly took him off.
